The rockers said they were ecstatic to be performing at the summer festival as they announced their 2007 European tour.
Townshend said: "When I woke up this morning one of the things I thought about is that I'm so happy to be able to say to my 17-year-old son (Joseph) that I'm headlining at Glastonbury.
"I'm 61 and it's the coolest thing to be able to say to him 'yes come down, come to Glastonbury, bring your friends and bring a caravan'." The Who World Tour will visit 14 countries and 28 cities in Europe.
It beings in Lisbon on May 16 and tickets go on sale from Monday. The dates are the first in the UK and continental Europe since the band's Endless Wire album came out last year, their first studio release in over two decades.
Townshend added: "It goes on and on and on and on and on. It gets better and better and better and better. It is more and more fun. But this chapter is over in July 2007 so if you want to catch the New Who at their most effective for 20 years, come and join us."
Daltrey, 62, said: "It's great to have been around for this length of time and still be playing new material, having it so well received and people liking the music. Life doesn't get any better than that."
The UK performances include Birmingham, Sheffield, Newcastle, Hull, the Isle of Man and two dates at Wembley Arena. There are also concerts in Swansea, Southampton, Cheshire, Dublin and Cork.
