Police and firefighters were called in the early hours of Sunday to a fire at a house in Tooting.
Five people from the same family — a 15-year-old girl, 16-year-old boy, 21-year-old woman, 22-year-old man and a 53-year-old woman — were badly hurt in the fire.
The 53-year-old mother and her eldest son escaped while the other three were rescued by firefighters.
All five were taken to hospital; four remain in "critical" condition, while the mother has been discharged.
Detectives are treating the fire as suspicious and traces of an accelerant have been found at the scene. A boy was arrested yesterday in connection with the investigation and taken to a south London police station where he was in custody.
Police are appealing for witnesses, including cyclists who took part in the London to Brighton cycle race on Sunday, to come forward.
Anyone with any information should call the incident room at Lewisham Police Station on 020 8721 4805 or Crimestoppers on 0800 555 111.
