Bloom, the 26-year-old star of Pirates Of The Caribbean and the Lord Of The Rings trilogy, managed to oust Prince Harry from the top spot thanks to his Hollywood good looks - according to Tatler magazine's judges.
The 19-year-old prince was last year's number one but this year comes in at number four.
Knightley, Bloom's co-star in the pirate tale, is just 18 years of age but is described as the "brightest young thing of her generation, with long limbs and fine features".
The list rates the 100 eligible men and women - chosen because they are unmarried (although they may be dating) and that they attend society functions. Other men in the top 10 include Sir Mick Jagger's 18-yearold son James, actor Hugh Grant and party organiser Ben Elliot.
Prince William makes the list of 100 men - "a serious hunk with a mind for art history" - but does not make the top 10.
Tatler editor Geordie Greig said sheer sex appeal put Bloom and Knightley at the top of the magazine's annual Little Black Book, adding: "It is a very dynamic and sexy list. Bloom is the name, face, body and mind whom all women would seem to feel happy having on their arm.
"Prince Harry has come into his own and he's become the social firecracker. He's got less responsibility than his brother, he's younger and slightly wilder."
Greig described James Jagger as "a shaven-haired mischief-maker who likes his rock chicks", while party animal Grant "gets better and better - no one is more witty and more intelligent".
The top 10 women include model Sophia Hesketh, Princess Anne's daughter Zara Phillips and Lady Tamara Grosvenor, daughter of the Duke of Westminster.
Greig added: "There's an amazing amount of incredibly good-looking people, and something very modern about the list."
But age is no barrier. Princess Diana's stepmother Raine Spencer, 73, is included in the list of 100 eligible women - while Jade Jagger and Gwyneth Paltrow miss the cut.
December's Tatler is on sale tomorrow.
