The victims - understood to be relations of Hollywood actress Helena Bonham Carter - were on a safari holiday when the vehicle they were travelling in lost control and overturned on Wednesday.
Three of the dead were named by sources as 14-year-old Eton schoolboy Marcus Egerton-Warburton, his grandmother Brenda Bonham Carter, 74 and step-grandfather Francis Kirkwood, 75.
A spokeswoman for the Foreign Office said: "We can confirm the deaths of four British nationals in a road traffic accident on August 20.
"We are still seeking clarification about the accident from the South African authorities."
At least two other members of the same family are believed to have sustained injuries in the crash.
Marcus's mother Fiona Bonham Carter is believed to have sustained a broken collar bone while his brother Piers received whiplash.
They were transported to Johannesburg hospital for treatment after the crash.
Reports suggest that the vehicle turned over after the driver struggled to keep control as a result of a blow out.
The four victims are believed to be part of the same extended family that also includes British actress Ms Bonham Carter.
