Air ambulance crews treated the man at the scene of the incident between junctions 13 and 12 on the M40 in Warwickshire, but he died.
It is thought the biker may have been a visitor to the Bulldog Bash at the nearby Long Marston airfield near Stratford - billed as Europe's number one biker party.
Both carriageways of the motorway in Warwickshire were closed between junctions 14 and 12 southbound and northbound between 10 and 13.
West Midlands Ambulance Service said they found a motorcyclist who had "suffered very serious injuries". He was pronounced dead at the scene.
The matter is now being dealt with by the police, he said.
Police confirmed they were treating the incident as murder and said an incident room has been set up at Warwickshire Police headquarters.
Detectives are appealing for information from motorists travelling on the M40 who may have witnessed the incident, said a spokesman.
Organisers of the Bulldog Bash said they were aware of a shooting.
A spokesman said: "At the moment we are all a bit shocked about the shooting - I've got nothing to say."
