Nearly 70% of party supporters in the capital want the flamboyant Henley MP to take on Labour incumbent Ken Livingstone in next year's election, a survey found.
His nearest rival appears to be ex-councillor Andrew Boff, who was backed by just under 10% in the research by grass-roots discussion site ConservativeHome.com.
The two other candidates - Warwick Lightfoot and Victoria Borwick - were the choice of 2.3% and 1.5% respectively.
The remainder of the 351 respondents said they were undecided or not planning to vote.
The hopefuls are due to go head to head at a series of hustings next month, and the winner will be announced at party conference on September 26.
Non-Tory members will be eligible to take part in the primary-style vote as long as they are on the London electoral roll, and register an interest.
There have been claims that Labour activists are planning to join the ballot in an effort to prevent Mr Johnson getting the nomination.
