The two boys, aged two and four, died in hospital after the blaze at the house on Rashcliffe Hill Road, in Huddersfield, West Yorkshire.
Police said the fire was not being treated as suspicious but investigations into its cause were continuing.
A spokesman said: "We are now working with fire investigators to establish the full circumstances of the fire."
Fire crews were called to the house at 1.12pm on Wednesday and, wearing breathing apparatus, they rescued the two young children from a bedroom.
The boys were taken by ambulance to Huddersfield Royal Infirmary, where they died later from their injuries.
Neighbours of the brothers said builders working on a nearby housing development had tried to rescue the children but could not save them.
An uncle and cousin of the victims' mother, who did not wish to be named, said they were "sad" about what had happened.
Other people said they were shocked to learn about the deaths of the boys, who were often seen playing with other children.
One neighbour, who did not wish to be named, said the mother of the victims has two other sons, believed to be aged five and six, and is pregnant with her fifth child. She said the Asian family had lived in the Housing Association property since it was built two years ago.
