Chris Hohn, a hedge fund manager who runs The Children's Investment Fund (TCI), donated £466 million to a foundation benefiting projects across the developing world.
The Hohns, who live in north London, have earned the titles of Britain's most generous philanthropists after giving nearly £800 million in just four years.
His donation dwarfs others in the UK, the Daily Telegraph reported.
Sir Tom Hunter, the Scottish billionaire, reportedly promised to give £1 billion to charity but that was spread over his life-time.
David and Heather Stevens, founders of the insurance company Admiral, gave away £100 million.
Mr Hohn set up TCI in 2003 so that a percentage of the earnings could go directly to its charitable arm, the Children's Investment Fund Foundation.
His wife, Jamie Cooper-Hohn, said she runs the charity using the business model of her husband's fund.
The foundation has given money to the Clinton Foundation for the treatment of HIV and Aids, to a scheme to help orphans in Malawi and to emergency aid projects in Darfur.
In an interview with the Sunday Telegraph, Mrs Cooper-Hohn said: "One of the things that attracted Chris and I was the shared sense of something that is larger than ourselves and the thrill that we can have a positive impact."
