Tony and Doreen Lofthouse, who made millions from the throat lozenges Fishermen's Friend, were confronted by burglars at their home near Blackpool, Lancashire, on Friday night.
The two robbers then made off with diamond-studded earrings, brooches and other high value jewellery as well as Mrs Lofthouse's MBE and OBE and a gold Fisherman's Friend lozenge which was mounted in a frame.
Lancashire Police said the couple, ranked 491st in the 2008 Sunday Times Rich List with a fortune of £165 million, were left in shock after being locked in a room.
Detective Inspector Glen Oldham, who is leading the inquiry, released a list of some of the stolen items and appealed for the public to be on the look out.
Mr Oldham described the robbery as "premeditated and carefully planned".
Police were called to the house, in Thornton Cleveleys, just before 8am on Saturday.
The robbery is believed to have taken place at around 9.30pm on Friday when the offenders, who had East European accents, forced their way inside and confronted the couple.
The Lofthouses, who are both in their 70s, were locked in a room while their home was searched.
Police believe the men fled the property about 10pm but it is not known in which direction they went or if any vehicle was used as part of their escape.
