The March4Justice group will deliver a petition with more than 30,000 signatures to Downing Street, with the document being handed over by five relatives of the dead, plus survivor Steve Hart.
The petition and marchers call for a police investigation into whether new criminal proceedings should be launched, to recognise that not all victims were dead by 3.15pm, and to release 300 boxes of information.
Former home secretary Jacqui Smith asked South Yorkshire Police to release the information 10 years early before she stepped down.
Andy Burnham, an Everton fan and former culture secretary, was key to the discussions about the information being released.
March organisers are hoping their presence in the capital will reapply pressure for their cause.
Organiser Barry Thompson, 39, is hoping for a crowd of about 5,000 to attend, but said: "The message on the petition is more important than the number marching.
"Alan Johnson is the new Home Secretary and Andy Burnham is the Health Secretary - these people have their own goals and aims and not necessarily those of their predecessors.
"We want to reaffirm that the Government still has an obligation to the families of the 96, the fans, the club and the city as a whole."
