Police called to a road accident discovered the man in the Land Rover. It had been involved in a head-on collision with a silver Ford Focus on Mackets Lane, Halewood, Liverpool, at 9.15pm on Saturday.
A Merseyside Police spokesman said: "When officers attended the scene, the driver of one of the vehicles, a gold-coloured Land Rover Discovery, was found to have suffered a head injury, thought to have been caused by a firearm. The driver of the Land Rover died at the scene."
The 36-year-old male driver of the Focus suffered whiplash and minor injuries and was treated by ambulance staff at the scene.
A Merseyside Police spokesman said: "The motive for the attack is unclear and it is not yet known who fired the shot towards the Land Rover."
The victim has not been formally identified but is believed to be a 35-year-old man from the Halewood area.
Police are carrying out forensic examinations around Mackets Lane, studying CCTV footage and making house to house inquiries. Detective Chief Inspector Mark Wadmore said: "We are carrying out extensive inquiries to build up a picture of events leading up to this incident and we are appealing to the public to come forward with any information which may assist us."
This shooting is not thought to be connected to any other firearms incident in the region, a Merseyside Police spokesman said. Mr Wadmore said: "Merseyside Police will not tolerate those who take firearms on to the streets and, be assured, our investigations will be thorough in pursuit of those responsible for taking this man's life."
Merseyside Police are also investigating another shooting in Liverpool on Saturday night. A 21-year-old man was shot in the leg in a "targeted attack" at a house in Gresham Street, Kensington.
Police believe several men forced their way into the property and shot the man in the leg at 9pm. They are believed to have left the area in a white Transit van. The victim required hospital treatment but his injuries were not life-threatening.
