Rhianna Clottey died from streptococcal pneumonia on Tuesday at Alder Hey hospital in Liverpool, a post-mortem examination revealed.
A spokesman for Alder Hey hospital, part of the Royal Liverpool Children's NHS Trust, said that it could not comment on the case because an internal investigation had been launched.
Merseyside Police confirmed they have launched an investigation into the youngster's death after they were contacted by the hospital.
A spokesman for the force said: "Merseyside Police's Liverpool North family crime investigation unit is investigating the circumstances surrounding the death of a four-year-old girl from the Old Swan area of Liverpool. The girl died suddenly in the early hours of Tuesday morning.
"A post-mortem revealed that she died of streptococcal pneumonia. It is a matter of routine that we always investigate sudden unexplained deaths."
No other details were released by the police.
A spokesman for Alder Hey said: "Our deepest sympathies are with the family at this sad time and our bereavement care team continues to support them.
"In keeping with our normal procedures, The Trust is carrying out its own internal investigation into the circumstances surrounding this death. The investigation is ongoing and we have no comment to make regarding the allegations."
A woman, who answered the door at Rhianna's family home in Shelton Close, Old Swan, said no comment would be made by the family.
