The officer was on duty in the Grosvenor Square area of central London when the accident happened. It is thought he was getting into a car when the weapon went off.
Scotland Yard said that the injured policeman was in a stable condition.
A spokeswoman said he had been a diplomatic protection officer, but stressed the incident had not been outside the US Embassy which dominates Grosvenor Square.
The shot was fired at around 10.10pm.
She said: "The officer shot himself in the leg. The incident occurred whilst he was on duty in a police vehicle.
"The officer was taken to a central London hospital where his condition is described as stable. Nobody else was injured."
She added that "as is routine" the Metropolitan Police's Directorate of Professional Standards was investigating the incident.
The matter has also been referred to the Independent Police Complaints Commission as is usual when a gun is discharged.
She added: "The officer will be relieved of operational firearm duties while the investigation is carried out."
