Chris Hawkins, 47, was arrested after Ryan was taken to hospital with serious injuries following an incident in Royd Street, Slaithwaite, Huddersfield, West Yorkshire, on Sunday.
The youngster, who had only recently celebrated his fourth birthday, died a short time later.
A 30-year-old man who was also detained has been released by police without charge but the youngster's father, who was arrested on suspicion of murder, remains in custody.
Ryan's 14-year-old sister Donna was also taken to Huddersfield Royal Infirmary with multiple stab wounds and was said to be in a serious but stable condition after surgery.
Detective Superintendent Tim Forber, of West Yorkshire Police, said the incident was not being treated as a random attack.
He said: "As you can imagine, his family are absolutely devastated by what has happened and they are being supported by specially trained family liaison officers."
Earlier, neighbours described how they rushed to Donna's aid as she shouted for help in the Pennine town where the ITV1 Sunday night drama Where The Heart Is was filmed.
Waqar Zaman, 34, owner of nearby Hill Top Stores, was one of the first people on the scene and described how he tried to help the teenager by removing his T-shirt and trying to stem the flow of blood from her wounds.
He said: "She had a stab wound on her face which went straight through her mouth, she had stab wounds in her stomach just under her rib cage, all over the place. Her right arm was completely broken, she couldn't breathe."
