James Oyebola died after being shot in the head and leg at close range in Fulham in July.
Rene McKoy, 19, and Kanyanta Mulenga, 22, have been remanded in custody after appearing at West London Magistrates' Court charged with murdering Mr Oyebola.
The Metropolitan Police said detectives have now also charged 23-year-old Dean Francis, of Roehampton, south west London, with the former British boxing champion's murder.
Mr Oyebola, who lived in north west London, was attacked at the Chateau 6 club in Fulham Road and died in hospital four days later.
After his death, his brother, Kelly, who runs a youth mentoring service, described him as a "gentle giant" and a "peacemaker".
Figures from boxing, including promoter Frank Maloney and former opponents, paid tribute to the 47-year-old.
The Oyebola family have since said they will establish a charity foundation in his name aimed at helping direct youngsters towards a brighter future.
McKoy, of Marmion Road, Clapham, south London, and Mulenga, of Tildesley Road, Putney, are due to appear at the Old Bailey on January 3.
Francis will appear at West London Magistrates' Court.
