The suspects, aged 21 and 20, and with links to Manchester street gangs, were arrested in separate prisons on Thursday.
They were questioned by detectives over the murder of the 15-year-old, shot dead in Broadfield Park, Moss Side, Manchester, in the early hours of September 9 last year.
Both men have now been bailed until September 20 and have been returned to prison, Greater Manchester Police said.
They were identified by an anonymous witness who came forward for the first time during the inquest into the teenager's death, which began on Monday.
The development, described as a significant breakthrough in the murder inquiry, dramatically halted the inquest so detectives could question the pair.
A spokeswoman for Greater Manchester Police said: "The two men arrested on suspicion of the murder of Jessie James have been bailed until Monday September 20 pending further inquiries.
"A 21-year-old man from Old Trafford and 20-year-old man from Hulme were arrested yesterday and have been returned to prison.
"A file of evidence has been passed to the Crown Prosecution Service for a final decision."
