A 27-year-old man, named locally as Gavin Battensby, was seriously injured when he tried in vain to save a toddler from a blaze at a house in Dufton Approach, Seacroft, Leeds, and died in hospital on Friday.
Mr Battensby, 15-month-old Kyra Moran and her mother were taken to St James's Hospital in Leeds but the little girl died soon afterwards, West Yorkshire Police said.
Mr Battensby died at Pinderfields Hospital, in Wakefield, where he was being treated at a specialist burns unit.
A 22-year-old woman was arrested on Thursday on suspicion of murder and remains in custody, a police spokesman confirmed.
He said: "Forensic inquiries are continuing and police are keen to speak to anyone who was in the area around the time of the fire to come forward."
Neighbours described how Mr Battensby tried to reach Kyra but was overcome by the smoke and flames. A friend said he ended up with 95% burns to his body.
Police and the fire service were called to the address just before 1.15am on Thursday.
Detective Superintendent Steve Payne said: "I would ask that anyone who may have been around Dufton Approach between 12.30am and 1.30am on Thursday to contact us as soon as possible.
"Do you live on Dufton Approach and may have looked out of the window between these times? Did you see anybody near to the house? A young baby has been tragically killed and we need the public's help to piece together what has happened."
