Di Santo has been picked in the squad for the tournament which begins in Venezuela on 19 January, but Scolari insists he is needed at Stamford Bridge.
The Chelsea boss, who has yet to start Di Santo in a game this season, is worried about his lack of cover for strikers Didier Drogba and Nicolas Anelka.
Argentina coach Diego Maradona is set to contact Chelsea to plead Di Santo's case, while the Argentine FA have turned to FIFA, who could order the club to let him go.
The 19-year-old claims even Chelsea captain John Terry is on his side and said: "I spoke to Terry a couple of times in the last couple of days and he said I was right to want to go.
"He said I'm only on the bench and need to get minutes in the team so I get noticed. I'm always on the bench.
"I spoke to Scolari and he told me that he wouldn't let me go."
