King made his first appearance of the season in the 5-1 win against managerless Fulham, with Spurs recovering after a poor start to the season.
Defeating Steve Coppell's side will move them 12th in the Barclays Premier League as they look to get into contention for a place in the top half in the second part of the season.
"We've been on a good run and we want to keep that going," said Spurs skipper King.
"Coming off a 5-1 win, we'll be confident but we know we have to do the right things to get a result against Reading."
Tom Huddlestone, who scored twice against Fulham, was delighted with the return of his captain following knee surgery in the summer.
"From his performance it seemed as if he'd hardly missed a game, despite being out for around six months," he said.
"He's one of the best centre-backs in the league and I think he proved that."
King's presence at the back should help head coach Juande Ramos continue to plug a leaky defence - but there are no problems at the other end of the pitch.
Robbie Keane's second goal in the win over Fulham took him to 100 Premier League goals in his career.
"It's great for me personally," said the Republic of Ireland striker. "I've joined Thierry Henry, Alan Shearer and Robbie Fowler on that list.
"It was Shearer and Fowler who I looked up to as a kid - to join them is very flattering, although you are never really aware of these things until you are told about them."
