The county's director of cricket, Steve Rhodes, has challenged the 21-year-old to fill the void left by the retirement of Graeme Hick.
Rhodes said: "Moeen Ali is a young and very talented player and is the kind of player we want to see flourish with the county."
He added: "He has already proved his worth but he has a great opportunity to cement a regular place in the team this year following Graeme Hick's retirement.
"I am delighted he has committed his future to Worcestershire."
Moeen said: "I'm very happy to commit to Worcestershire and repay the faith that the club, and Steve Rhodes in particular, have shown in me.
"New Road is an exciting place to be at the moment and I'm delighted to be part of a team that I believe can do well in all formats of the game."
Moeen made the move across the Midlands from Warwickshire for the 2007 campaign and played a key part in Worcestershire winning the Pro-40 Division One title - their first major honour for 13 years.
His opening partnership with Steven Davies of 151 in just 17 overs helped secure the title in the deciding game against Gloucestershire at Bristol.
Now Hick's decision to quit New Road after 25 seasons will give Ali with the chance to establish himself in the middle order in the county's LV County Championship line-up after gaining promotion from Division Two in 2008.
