Matthew Hayden was unbeaten on 77 alongside Michael Hussey, who had reached 43 not out at the break.
On the third day, the tourists had completed their first innings 532 all out in response to Australia's 463.
And having resumed 13 without loss in their second innings, Australia added 77 by lunch and Hayden and Hussey continued their stand after the break as the hosts chase a world record-equalling 16th consecutive Test victory after winning the series opener in Melbourne last week by 337 runs.
In the morning session, Anil Kumble claimed his 100th wicket against Australia when his delivery was hit square by Phil Jaques (42) straight to Yuvraj Singh.
And Ricky Ponting's wicket soon followed when he was caught out by VVS Laxman from a Harbhajan Singh delivery, prompting wild celebrations from the 27-year-old.
