Fifth seed Kuznetsova became the highest-profile casualty of the first week of the championships as Lisicki claimed a 6-2 7-5 victory on Court One.
Saturday's other big shock saw teenage rising star Melanie Oudin stun Jelena Jankovic with a brilliant fightback to knock the sixth seed out of Wimbledon.
Oudin, a 17-year-old from the state of Georgia in the United States, came through qualifying to reach the main draw but is now heading for the last 16 and making a name for herself.
She dropped the first set on a tie-break to Serbian Jankovic, a former world number one, but fought back to win 6-7 (10/8) 7-5 6-2.
Defending champion Venus Williams had little trouble seeing off Carla Suarez Navarro in their third-round match on Centre Court.
In their only previous meeting, in the second round of the Australian Open earlier this year, the Spaniard triumphed in three sets, but it was a different story at SW19 as Williams eased to a 6-0 6-4 victory.
Earlier on Court Two Ana Ivanovic demonstrated some of the form which made her world number one as she booked her place in the fourth round at Wimbledon as she beat Australia's French Open semi-finalist Samantha Stosur 7-5 6-2.
Top seed Dinara Safina also cruised into the fourth round at Wimbledon with a 7-5 6-1 victory over Belgian Kirsten Flipkens.
