The Nottinghamshire seamer struck with the ninth delivery of the match - which had already been reduced to 43 overs a side - to remove opener Herschelle Gibbs to boost England's hopes of securing the victory which would propel them to second in the world rankings.
Broad tempted Gibbs into driving outside off-stump and wicketkeeper Matt Prior reacted impressively to dive in front of first slip to take a superb one-handed catch and justify captain Kevin Pietersen's decision to bowl first after winning the toss.
But England were only able to deliver one more over before further showers, which have been forecast all day at the SWALEC Stadium, halted play again with South Africa on six for one after three overs.
England had earlier decided to name an unchanged line-up for the fourth successive match after all-rounder Andrew Flintoff shrugged off the back spasm which had troubled him during Sunday's win at Lord's.
South Africa, desperately seeking to end their losing run, made one change with batsman Justin Ontong making his first appearance of the series as a replacement for all-rounder Vernon Philander.
