The former Australia captain sustained the injury in Friday's defeat to Wigan and scans confirmed the break.
With Leeds currently second in the table and the Grand Final 11 weeks away, Buderus faces an anxious wait to see if he can play again before the season's climax.
Rhinos physio Meirion Jones said: "Fortunately we were able to get Danny in for an X-ray on Saturday morning and to see our specialist but unfortunately he has confirmed that Danny has suffered a fracture to his tibia.
"He will be in plaster for six weeks and then will begin a period of rehabilitation after that, which we will assess the length of once he has seen the specialist when the plaster is removed."
Head coach Brian McClennan added: "We are all disappointed for Danny, especially as he has been playing so well. However, he will receive the best possible treatment from our medical staff and they will have him back as soon as he is fully fit.
"We have got him seen by the specialist at the earliest opportunity and now the hard work starts for Danny to get back to action.
"Every team suffers injuries during the course of a season which is why you need a strong squad, and we have every confidence as a group that we can continue to build towards the play-offs starting next Saturday against Warrington."
