The former drugs cheat needed to finish first or second in Birmingham to stand any chance of selection for the Beijing Games, for which he is currently banned by the British Olympic Association.
The 30-year-old won in a time of 10.00 seconds, meaning his fate now lies in the hands of a High Court judge, who will decide on Wednesday whether to issue an injunction against his ban.
"That was hard work. I'm just glad I've done my part of it," Chambers said on BBC1. "Hopefully, next week now things will go well."
Chambers finished ahead of second-placed Simeon Williamson, who secured his Olympic place with a time of 10.03.
Chambers said of the race: "It was tough. I tried to keep my head clear. My biggest fear was messing up.
"I thought, 'Keep cool, keep cool'. I'm so happy."
Asked how he would cope with the impending court case, he added: "It's going to be tough but I'm going to keep my spirits high.
"I'd like to say thank-you for everyone that supported me throughout this whole ordeal.
"Hopefully, all that will go in my favour for Beijing this summer. I just want to go and do well for my country."
