The Arsenal left-back was carried off on a stretcher early in the second half of the 3-0 win over Estonia, with victory at Wembley meaning England will qualify for Euro 2008 if they win in Moscow.
"He rocked his ankle blocking a shot," said England boss Steve McClaren. "It doesn't look good but injuries can settle down in 24 hours and it is a critical period. We'll know more on Sunday evening when we reassemble. They have X-ray machines here so we're waiting for the results."
McClaren brought on Joleon Lescott as a half-time substitute and he went to left-back following Cole's injury, and Nicky Shorey is also in the squad if the Chelsea star does not make it.
John Terry missed the win with a knee injury but has not been ruled out of the Russia clash.
"He is a lot better and a lot more positive than he was on Thursday," McClaren added. "He's another one where 24 hours can be a long time for injuries and we're just hoping that come Monday he'll be able to train."
