Drew, who captained the side to their win over Bradford last Sunday in the absence of Chris Thorman, looks set to join Wakefield, while Nero - who is out of contract at the end of the season - has been linked with a move to Bradford.
Giants managing director Richard Thewlis said: "Such has been the media speculation surrounding our players that we felt it appropriate to formalise this news now rather than at the season's end, so that we can all keep our focus on making the top six."
He added: "It will also give all our supporters the opportunity to turn out in force at our final scheduled home game when we meet the Wakefield Wildcats and give these players the send-off they deserve."
Hooker or half-back Drew had another year left on his contract, but the club have decided not to stand in his way over a move.
"I have had a great time at the club - but now is the time for me to move on," he said.
"I would like to thank everyone at the Giants for giving me the chance to play in the Super League, and to the fans for giving me such great support. I wish them all the very best for the future.
"Like the club, my focus at the moment is solely on achieving a top-six play-off place - and then who knows how far we could go?"
Thirty-one-year-old Reilly, Huddersfield's longest-serving player, had hoped for a new contract after reversing his decision to retire - and is now likely to look for another club.
He said: "The progression of the club has been almost unbelievable, and my memories of the players and games I have played in will never fade."
