He becomes the latest Scotland international to return home, following Chris Cusiter and Scott MacLeod's moves to Glasgow and Edinburgh respectively.
He will return to his homeland from Ulster this summer after agreeing a three-year deal.
Dewey, who spent two years with Edinburgh, said: "I'm looking forward to returning to Scotland and joining Glasgow.
"Glasgow have been on the up for the last three seasons now and their win over Toulouse in France back in January really shows they are a team to be reckoned with.
"It's too good an opportunity to miss out on.
"It's going to be tough to earn a place in a very competitive Glasgow back-line, however, I'm looking forward to the challenge."
Glasgow head coach Sean Lineen added: "I've always liked the way Rob plays. He has a very big physical presence.
"With Chris Cusiter joining the club in the summer and Thom and Max Evans re-signing until after the next World Cup, Rob is yet another exciting addition to a pretty special looking Glasgow back-line."
