The 26-year-old former Sydney Roosters and Manly half-back scored 32 tries in 50 appearances for the Quins before joining Salford in 2007 and has spent this season with Castleford where he is their leading tryscorer with 19 touchdowns.
Dorn, who will make his final appearance for the Tigers against Bradford on Saturday, said: "I am really excited about coming back to London. I am very much looking forward to working with Brian McDermott again and with Billy Peden. They are two coaches who are very good at getting the best out of their players."
Harlequins coach Brian McDermott said: "I think everyone recognises Luke as one of the most potent attacking players in Super League. The squad will benefit from his style of play."
Dorn joins fellow Australians Luke Williamson (Manly Sea Eagles) and Chad Robinson (Parramatta Eels) who have also signed on for two years at the Stoop from 2009.
