Replying to 264 for eight after Guyana-born Chanderpaul returned to his roots to hit an unbeaten 112 off 126 balls, the tourists' hopes of claiming successive triumphs were undermined by the loss of two wickets inside the first eight overs.
They lost opener Ravi Bopara and Kevin Pietersen in a three-over spell and slipped to 36 for two after both played on.
But a determined stand between captain Andrew Strauss and Owais Shah helped them recover and reach 73 for two after 15 overs.
England had only themselves to blame for facing such a challenging target, having also claimed two early wickets but then dropped Chanderpaul on 27 when West Indies were rebuilding the innings on 80 for two.
Lancashire seamer Jimmy Anderson gave England a flying start by removing both openers during a new ball spell of 6-1-15-2 which left West Indies 24 for two.
But Chanderpaul, returning to his home island, repaired the early damage in a brilliantly well-paced 133-run stand with Ramnaresh Sarwan (74) which should have set up West Indies for an even bigger total.
