Left-arm spinner Panesar claimed five for 46, including three in 24 deliveries to decimate the West Indian middle-order - they finished 222 all out - and leave England with a chase of 110 to complete the success.
Captain Michael Vaughan and star batsman Kevin Pietersen then combined as the hosts charged to a seven-wicket win.
Vaughan finished unbeaten on 48 while Pietersen hit 28, having come together after Daren Powell accounted for openers Alastair Cook and Andrew Strauss inside eight overs.
Things appeared to be drifting away slightly after lunch but a Panesar-inspired England took the final six wickets at a cost of 60 runs to set up a comfortable pursuit.
Fast bowler Steve Harmison weighed in with a couple of wickets, and sent down a prolonged 17-over spell of hostility despite confirmation he will need surgery on a hernia problem.
Once again Shivnarine Chanderpaul defied England's push in tandem with last man Corey Collymore, before perishing after 17 hours and 40 minutes undefeated across three innings.
Chanderpaul was finally dismissed when he was bowled aiming a slog-sweep at Panesar, having top-scored with 70.
A rush of blood from all-rounder Dwayne Bravo began the West Indian demise as, immediately after lofting a straight four, he mishit another blow down the ground off Panesar.
Two classic turning deliveries then accounted for Marlon Samuels and Denesh Ramdin, the first edging to slip and the second bowled by one which pitched on middle stump and hit off. Harmison pitched in with an action-replay dismissal of Powell, who was caught at mid-off hooking at a short one, while Fidel Edwards had his stumps rearranged.
