Ennis won the opening two events in the Olympic Stadium, clocking 12.93 seconds in the 100metre hurdles and clearing 1.92m in the high jump.
Olympic champion Nataliya Dobrynska then looked set to make huge inroads into Ennis' lead with a winning effort of 15.82m, with Ennis only managing 13.07m in the opening round and 12.55m in the second but Ennis dug deep to produce a personal best of 14.14m with her final attempt for a total of 3,070 points, 138 ahead of Dobrynska in second place.
Britain's Louise Hazel was down in 21st place after 11.62m in the shot.
In the men's 1,500m, Andy Baddeley and James Brewer advanced to the semi-finals but team-mate Tom Lancashire missed out.
Baddeley was fourth in his heat in three minutes 45.23 seconds and Brewer, 21, ran a mature tactical race in his first senior championships to set a new personal best of 3mins 37.17secs in finishing third.
Brewer was a late addition to the team and said: "I think I have proved out there I deserve to be here. I stayed on the inside and when I got the opportunity I moved out and it was almost perfect."
Lancashire was well placed in his heat with 100m to go but could not produce a sprint finish and finished eighth.
