The 27-year-old former Auckland Warrior, who has featured in the top five in the list of Super League tryscorers in each of his three seasons with Warrington, has agreed a two-year deal with the Londoners.
"We're very excited about having a proven tryscorer," said coach Brian McDermott. "He's been either the top or one of the top tryscorers in Super League over the last few years. He can play back row as well as centre and wing. It's great that he's keen to come down here and further his career."
Fa'afili, who has scored 20 tries so far this year but is being squeezed out of Warrington by the salary cap, said: "It's going to be a new challenge for myself and I'm looking forward to it."
Meanwhile, Harlequins captain Rob Purdham will miss his club's last two matches after suffering a serious shoulder injury in Friday's defeat at St Helens.
The England skipper, who was out for two months in the middle of the season with a back problem, sustained his latest injury 15 minutes into the game against the champions and was unable to return.
Purdham will undergo surgery on the same left shoulder for the second time on Monday, ruling him out of contention for a Great Britain call-up for the Test series against New Zealand.
"It's the end of what has been an unlucky season for Robert," said McDermott.
"He is an invaluable captain who leads by actions and not words. He will be a great loss to a team that looks up to him."
Full-back Mark McLinden, who has been cleared of a broken toe, will lead the side against Salford in Saturday's final home game of the season if he passes a late fitness test.
