Federer, who has been fighting a stomach virus since his arrival in Australia last week, is aiming to become the first man in the Open era to win three consecutive Australian Open crowns.
The top seed could face Fernando Gonzalez, his opponent in last year's final, in the last eight.
Argentina's David Nalbandian has been drawn against Australian wild card Robert Smeets, while local favourite Lleyton Hewitt, the 19th seed, will meet Belgium's Steve Darcis and American James Blake will take on Chile's Nicolas Massu.
One of the highlights of the first round will be the clash between two former finalists - Sweden's Thomas Johansson and 15th seed Marcos Baghdatis.
The winner of that match is likely to take on the dangerous Marat Safin - winner at Melbourne Park in 2005 - in the second round.
