The 31-year-old Lancashire all-rounder sustained the injury during the third Test in Antigua over a month ago and has been having intensive treatment on it ever since, flying home for a week before returning to join the squad at the start of this series.
"I've bowled as near to 100% as I probably can do in a net because playing in a match brings a bit more out of you," said Flintoff.
He added: "I've been bowling for the last week, bowled 10 overs over the previous two days and done my training and work. I was close to playing on Sunday but we erred on the side of caution and I'll definitely be fit for Friday."
Flintoff was close to selection for Sunday's 21-run defeat in Guyana, which levelled the five-match series at 1-1, but England decided to be cautious and will welcome him back at the Kensington Oval on Friday.
England now face a selection headache about who will make way for their talisman with James Anderson and Steve Harmison both excelling in Sunday's match while Stuart Broad was impressive in Friday's opener.
The tourists could decide to operate without a spinner to accommodate the return of Flintoff, which would mean Gareth Batty dropping out on Friday.
