West Indies captain Gayle, the only man with a 20-over international hundred to his name, hit 88, including six monstrous sixes, as his team got home with 25 balls to spare.
His half-century came from just 23 balls, which outpaced his opening partner Andre Fletcher, whose own came from 31.
Both men were out to skied catches after racing to 121 without loss by the halfway stage of their chase of 170.
Earlier, Australia were restricted to 169 for seven after they won the toss and decided to bat first.
Ricky Ponting's team slumped to 15 for three inside four overs but they were able to overcome such a start and reach a respectable score largely due to a determined half-century from opener David Warner, whose 63 off 53 balls ensured Australia did not completely capitulate.
Warner found an ally in wicketkeeper Brad Haddin, who helped Australia recover with a 66-run stand which allowed their middle order to play with a greater expression during the closing overs.
