The left-hander was put down on four and 20 and looked out of touch but somehow survived to help his side to 69 for one after 15 overs.
Gayle decided to bat after winning the toss and play was able to start on time despite some parts of the outfield still being soggy after the recent heavy rain which had left the ground flooded on Thursday.
He was twice beaten outside off stump in the opening over from James Anderson, who was fined half his match fee in the previous game at Edgbaston for barging into Windies batsman Runako Morton.
Anderson's new ball partner Liam Plunkett began with two legside wides and saw Devon Smith drive his first legitimate delivery to the off-side boundary and then despatch a wide half-volley for another four.
But Anderson made the initial breakthrough for Paul Collingwood's side in the third over with 16 on the board.
Smith picked up his third boundary with another scorching off-drive but he cut the next delivery to Ian Bell at backward point after making 13 off nine balls.
His dismissal brought in the prolific Shivnarine Chanderpaul, who passed a late fitness test on the ankle injury sustained making an unbeaten 116 at Edgbaston.
Gayle got off the mark in the fourth over with a square drive off Plunkett but the Durham bowler could have had immediate revenge.
The left-hander smashed the next delivery straight at Alastair Cook at short extra-cover but he was unable to hold on to the difficult chance.
