The announcement was made in Colombo, Sri Lanka, where member countries of the Commonwealth Games Federation gathered to hear final presentations before submitting their votes.
Scotland's First Minister Alex Salmond hailed Glasgow's victory on BBC1 Scotland and pledged: "We will make these Games the greatest sporting event our country has ever seen. They will be our chance to show the whole world the very best of Scotland."
It will be the first time Scotland's largest city has staged the Games, although the capital Edinburgh has had it twice - in 1970 and 1986.
The Glasgow bid won by 47 votes to 24 and members of Glasgow's team had visited 67 of the Commonwealth's member nations in a charm offensive designed to secure votes.
The Nigerians had concentrated on the emotional appeal of securing what would have been the first Commonwealth Games to be held in Africa.
Sportscotland chair Julia Bracewell was delighted by the Glasgow victory and predicted the result would have a lasting impact on the country.
She said: "It is fantastic news that Glasgow has won the right to host the 2014 Commonwealth Games for Scotland.
"Hosting such a high-profile event will undoubtedly help to drive sport forward at all levels, increasing participation and improving performance.
"It will provide Scottish athletes with the advantage of competing and being supported on home soil. And as importantly, it will give coaches, officials and volunteers the experience of a lifetime."
