Saeed bin Suroor's second string, ridden by Ted Durcan, led home stablemate Kite Wood, the mount of Frankie Dettori.
Godolphin were securing a first domestic Group One victory of the season and it was also the first time Sheikh Mohammed's team had tasted British Classic glory since Rule Of Law won the Leger five years ago.
Kite Wood (9-4 favourite) went clear of the pacemaking Von Jawlensky as the field bunched approaching the two-furlong marker, but Mastery (14-1) came to tackle him.
Durcan's mount got on top in the final 50 yards and scored by three-quarters of a length, with Monitor Closely, who was supplemented for the race to the tune of £45,000, taking third, another length and three-quarters back.
Durcan said: "Sheikh Mohammed and everyone have been a huge help to me over the years. This horse has loved the ground and stays really well."
