While West Brom lost 4-2 at home to Coventry, Adrian Boothroyd's side found themselves two adrift early on after Clive Platt and Matthew Connolly scored for Colchester inside the first 28 minutes.
But Marlon King pulled one back and John-Joe O'Toole equalised before the break - and the comeback was complete when Tommy Smith's delightful chipped cross left Tamas Priskin with a simple header to collect all three points and end their poor recent run.
At The Hawthorns, Leon Best and Michael Mifsud silenced the home crowd as Coventry took a 2-0 lead.
But left-back Paul Robinson flighted a perfect cross for Roman Bednar to head home and drag West Brom back into it, before Chris Brunt fired in an equaliser.
Robinson's night took a turn for the worse, however, when he was sent off for a second caution and Coventry had a late Mifsud goal before Best and Mifsud netted in the last seven minutes for the Sky Blues.
Third-placed Wolves lost ground on the top two with a 1-0 defeat at Barnsley.
After a goalless first half Istvan Ferenczi put the Tykes ahead, only for Wolves to win a penalty soon after. However, Andy Keogh's kick was saved well by Heinz Muller and Mick McCarthy's side were unable to force their way back.
Matt Holland and Andy Reid helped Charlton eke out a 2-0 win at Cardiff to move third while Bristol City maintained their early-season promotion push with a 2-0 win at home to a poor Ipswich side at Ashton Gate.
City went ahead through Marvin Elliott's fifth-minute thunderbolt and Bradley Orr's penalty - after a handball by Alex Bruce - made Ipswich's task all the more difficult.
