The 20-year-old was injured during the Barclays Premiership win over Reading on Sunday after coming on as a second-half substitute.
He will miss the UEFA Cup quarter-final tie against Sevilla and will still be a doubt if Spurs beat the La Liga high-flyers and reach the last four.
"He has injured his knee and could be out for three or more weeks and that is a blow," said Spurs boss Martin Jol.
Huddlestone, an England Under-21 international, has established himself as a player of promise at White Hart Lane this season, although he has recently been on the bench following the return of Jermaine Jenas from injury.
In general, Jol has been fortunate with injuries in his squad and their recent good form has taken them back into the top six of the Premiership.
They have been without skipper Ledley King at centre-back since Boxing Day and Huddlestone was among the players who could drop into defence.
With Anthony Gardner injured with a fractured leg and Ricardo Rocha cup-tied, Pascal Chimbonda is likely to be moved from full-back for the first leg against Sevilla on Thursday.
