The Indian captain shared an eighth wicket partnership of 107 with Harbhajan Singh and an unbeaten final wicket stand of 57 with Ishant Sharma to put the tourists well in control of the final game of the four-match series.
Kumble and Harbhajan came together before lunch with the Indian total at 359 for seven following the dismissal of Sachin Tendulkar for 153 and frustrated the Australian attack for 35 overs.
Harbhajan, making his first appearance for India since the controversial second Test in Sydney, recorded his fourth test half century, hitting seven boundaries in a knock of 63.
Having survived a loud lbw appeal by Hogg before lunch while he was on 13, Harbhajan was finally dismissed when he attempted to slog an Andrew Symonds delivery over midwicket but got a top edge that flew high into the air and was taken at square leg by wicket keeper Adam Gilchrist.
RP Singh went six balls later for a duck, lofting a ball by Michael Clarke to Mitchell Johnson at midwicket, but Kumble and last man Sharma continued to frustrate the Australians as they took the Indian total past the 500 mark.
Tendulkar had earlier made 150 for the 17th time in his Test career after India resumed at 309 for five.
The tourists added 27 runs in the first three and a half overs of the day before MS Dhoni was dismissed for 16. The Indian wicketkeeper attempted to cut a Mitchell Johnson delivery to the boundary but was caught at deep point by Andrew Symonds.
Tendulkar reached the 150 mark with his 13th four of the innings but required treatment after Lee's next delivery when he got an inside edge to the ball which struck him on the inside of the knee.
The veteran then attempted to pull Lee's next ball but put his shot high to deep square leg where Brad Hogg took a simple catch to dismiss Tendulkar.
