Kallis gave South Africa an injury scare when he suffered a knock to his finger during the Proteas' warm-up match with Bangladesh 'A' at New Road on Thursday and retired injured on 55 during his side's second innings.
But the former Middlesex player has allayed any fears about his fitness by coming through a full training session in Birmingham on Monday morning.
A South Africa spokesman said: "Jacques is 100% fine. He has been in the nets today (Monday) at Edgbaston and taken part in a full practice session."
South Africa will be looking for Kallis to finally show the form that has made him one of the top batsmen in the world.
Although he has contributed with the ball during the current series, he has managed to score only 24 runs in three innings with a highest score of 13.
South Africa opener Neil McKenzie will win his 50th Test cap at Edgbaston. The 32-year-old has scored 2,848 runs at an average of 39.55 in the five-day form of the game.
Paceman Andre Nel is expected to make his first Test appearance since January in place of the injured Dale Steyn.
