The second leg at Eastlands will mean a return to England for the Bundesliga side`s former Tottenham boss Martin Jol.
The other last-eight clashes will see Paris St German take on Dynamo Kiev, Shakhtar Donetsk play Marseille and Werder Bremen challenge Udinese.
Hamburg will be a step up in class for Mark Hughes' men, who needed penalties to scrape past Aalborg in the last round.
The Bundesliga outfit came from 2-0 down to beat Galatasaray 3-2 in Istanbul on Thursday night and are very much in the race for the German title.
The two French sides left in the competition both face eastern European opposition, with PSG hosting Kiev and Marseille travelling to Shakhtar in the first legs of their last-eight clashes.
Udinese, who travel to another German side, Bremen, are the last Italian club left in European competition this season.
A north German derby could be on the cards in the semi-finals, with Hamburg and Bremen due to meet if they can get past City and Udinese respectively.
The winners of Bremen and Udinese will be at home for the first leg. The other last-four clash could be either an all-Ukrainian or all-French affair, with the winner of PSG and Kiev facing either Shakhtar or Marseille.
The first leg of the quarter-finals will be played on April 9 and the second on April 16. The semis will take place on April 30 and May 7, with the final on May 20 in Istanbul.
