The 28-year-old made a tournament-low eight-under-par 64 on Friday as 47 players were left on the course after a two-and-a-quarter-hours delay following high winds at Pinx Golf Club, Jeju Island.
McDowell, from Portrush, exploited the best of the conditions as the players fought frantically to avoid an early wake up, with Johan Edfors, Jeev Milkha Singh, Thomas Bjorn and Padraig Harrington all completing their rounds in near darkness and making an impact at the top end of the leaderboard.
Sweden's Edfors and India's Singh were joint second on 10 under, while Denmark's Bjorn and Ireland's Harrington were on eight under, alongside American Anthony Kim, who carded a second successive 68 earlier in the day.
Ireland's Paul McGinley and Kim Hyung-tae were three shots behind, while fellow South Korean Hwang Inn-choon finished off a round of 69 today to move to seven under overall.
Finland's Mikko Ilonen, who led following an opening round 67, claimed an eagle at the 9th, his last, to join Hwang at seven under alongside Australian Terry Pilkadaris.
Japan's Shingo Katayama completed a round of 70 to move to six under alongside American Chris DiMarco and Ariel Canete, of Argentina.
Northern Ireland's Rory McIlroy missed the cut, which was made at level par, alongside Scottish duo Simon Yates and Stephen Gallacher, who finished on one over and two over, respectively.
The third round is set to begin between 10.30am (0030 GMT) and 11am local time.
