Murray defeated world number one Roger Federer in three sets on Monday but Verdasco, ranked 19 places below him, proved a resilient opponent.
The British number one was able to come out on top in a tie-break in the deciding set to claim a 6-3 3-6 7-6 (7-5) victory in a gruelling contest at the Aviation Club Tennis Centre which lasted two hours and 25 minutes.
The world number 11 earned his first break of the match to go 4-2 ahead in the first set, and the Scot, who won both of the pair's previous two meetings in St Petersburg and at the Australian Open, coolly wrapped up it up 6-3 with his third ace of the match.
The world number 30 responded in positive fashion and produced a love service game in the first game of the second set before a feeble Murray drop shot allowed Verdasco to break his serve and go 2-0 up.
Murray missed a chance to break as the 24-year-old from Madrid saved three break points to hold serve, take a three-game lead and go on to claim the set.
Serve dominated in the deciding set which went to a tie-break. Murray took an early lead in the breaker and went 3-0 up after he served first before a solid volley from Verdasco saw him pull one back.
Murray moved ahead with serve to take four match points after Verdasco's mistakes handed him the initiative.
The first three went begging with terrible forehands, but a 34-stroke final rally ended with Verdasco going long with an unforced error to give Murray victory.
The Scot will now face world number five Nikolay Davydenko in the next round after the 26-year-old Russian eased to a 6-1 6-4 victory over the Czech Republic's Jan Hernych.
