The 25-year-old loose forward made an abortive comeback six weeks ago following a broken shoulder blade he broke in May and scans have now revealed the bone has still not fully healed.
Wigan coach Brian Noble, who had hoped to have his captain back for the Super League play-offs, told a press conference: "This is a serious blow for us and for the player."
He added: "It's a shame for the lad but clearly he now needs to rest it for at least four weeks before he can start training again.
"He will not be ready for the World Cup which is another blow for England and for him.
"However, it is important not to take any risks and to have him fully fit for the start of the new season."
O'Loughlin played in all three of Great Britain's Tests against New Zealand last year but missed England's mid-season international against France because of his injury.
He is the second player to be ruled out of England coach Tony Smith's World Cup plans following the decision of Bradford forward Sam Burgess to undergo shoulder surgery.
