The 2004 runner-up set up a quarter-final against Adrian Lewis thanks to a 4-2 victory at Alexandra Palace, which he very nearly sealed with a nine-darter.
The contest appeared to be going Van Barneveld's way when he took the first set, but the match soon turned as Painter won six of the next seven legs to go 2-1 up. Van Barneveld fought back with equal determination, reeling off four legs to edge ahead.
The fifth set went with throw until a 180 from Painter allowed him to move 3-2 ahead overall.
It looked certain to go 3-3 when Van Barneveld raced to a 2-0 lead in the sixth set but Painter came roaring back again, eventually sealing victory with a 10-darter after landing the first seven darts of a nine-dart finish.
Painter's next opponent Lewis survived a titanic battle with Tony Eccles earlier in the day.
Eccles came from three sets down to defeat Steve Maish on Thursday, but was on the receiving end of a comeback as Lewis came from 2-0 down in sets and 2-0 in legs to take a 4-3 win in the decider.
Peter Manley ended Jan van der Rassel's giant-killing spree with a 4-1 victory. Van der Rassel had defeated Colin Lloyd and Denis Ovens in the opening two rounds, but found the three-time finalist too much of an obstacle to overcome in the last 16.
Manley will face another surprise package in the quarter-final in the shape of Kirk Shepherd, who continued his superb debut run by beating world number 13 Barrie Bates 4-3.
James Wade overcame a bout of flu to beat Mark Dudbridge 4-2. Wade will play 2003 champion John Part in the last eight after the Canadian whitewashed Alex Roy 4-0 in the final match of the day.
