The new deal adds an extra 12 months to Peacock's previous contract - which still had two years to run - and will keep him at his hometown club until the end of the 2011 season, when he will be approaching 35.
Peacock, 31, who joined the Rhinos from Bradford in 2006, has also been granted a three-month testimonial by the Rugby Football League for services to the game.
The prop, who is in Jacksonville as part of the Rhinos' pre-season training camp, said: "I am delighted to have agreed the new deal and I am looking forward to the season ahead immensely.
"The England players only began pre-season at the start of January, so the trip to Florida has given us a chance to get back together with the other guys and turn our thoughts to next season and the challenges ahead.
"I would like to thank the Rugby Football League for granting me a testimonial, and it is a huge honour.
"I am getting together my testimonial committee. But with Jamie Jones-Buchanan also enjoying a well-earned testimonial this year, I do not want to hold any events that will clash with his plans - so that is paramount in any arrangements we make."
The news is a timely boost for the Rhinos as they prepare to defend both their Super League title and their World Club Challenge crown against Manly on March 1.
Leeds coach Brian McClennan added: "Jamie is a world-class player and a world-class guy. I am delighted he has agreed a new long-term contract and I am sure that we are going to see him go from strength to strength in the coming years.
"We have a nucleus of key players now who have committed their long-term future to the club, and that helps us continue to build the spirit and personality of the team and the club."
