Two assailants reportedly used a dump truck to ram their way into the police station in Kashi and then threw two hand grenades.
A further 16 people were injured, while the attackers have been arrested.
The attack came just four days before Friday's Olympic Games opening ceremony in Beijing.
Police have reportedly identified five terrorist groups in the region who were allegedly plotting to sabotage the Games, according to Xinhua.
The Xinjiang Uygur Autonomous Region, which has been the heart of China's security fears, is home to a Sunni Muslim ethnic minority.
A team of policemen were undertaking morning exercise in Kashi at about 8am local time when the attack took place.
Fourteen policemen were killed at the scene, while two others died on their way to hospital.
Police had information suggesting that the "East Turkistan Islamic Movement" planned to make terrorist attacks in the week prior to the Games, according to Xinhua.
